Summary
Spot blotch (SB) caused by Cochliobolus sativus has been the major yield-reducing factor for barley production during the last decades. Unlike other proteomic research on plant response to pathogens, this study was focused on the proteome response of SB adjacent to the infected areas in resistant and susceptible genotypes. Replicated two-dimensional polyacrylamide gel electrophoresis experiments were conducted to detect proteins differentially expressed under inoculated and non-inoculated conditions. Twenty out of the 200 identified proteins showed significant variation in relative abundance after pathogen challenge of the resistant and susceptible genotypes. After tryptic digestion, MALDI-TOF/MS analysis and MASCOT database searching identified proteins with presumed functions relating to signal transduction, proteins involved in energy, general metabolism and protein synthesis. All of the identified proteins have been previously reported as being involved in disease response which suggests a common baseline defense strategy for different host-parasite interactions.
